In order to study the relationship between food preference and brain function, we conducted experiments on fMRI and fNIRS for healthy volunteers. The sensitivity of the smell is more significant in women, and the function decreases in both sexes in age dependence. FMRI experiments show that the sex difference and age-dependent functional changes are particularly involved in the functional connectivity of the frontal medial cortex and angular gyrus. In addition, a study using fNIRS suggested that there may be differences in frontal lobe function at the time of eating depending on the living environment (whether it is with the family or alone) and the preference of the usual food (whether they like the salty taste or the mild taste). In the future, we plan to expand the number of subjects to continue the fNIRS experiment.

周産期のストレスが脳の正常な発達を妨げ、うつ病を代表とする精神疾患の罹患率を上げることが知られている。このような変化は動物実験においても再現されているが、その背景にあるメカニズムは未解明である。 研究代表者らはこれまで、母仔乖離マウス(Maternal deprivationマウス; MDマウス)を用いた研究を通じて、周産期ストレスがグルタミン酸恒常性の破綻やシナプスの不安定化を引き起すことを示してきた(Takatsuru et al., 2009, Toya et al., 2014ほか)。その後の研究で、初老期のMDマウスでは認知力が著名に低下していることも発見した。この研究では若年期の認知能力は正常であったが、これまでの知見から、MDマウスは特に性成熟前期(6-8週齢)にシナプスの脆弱性が顕著であることを報告しており(Takatsuru et al., 2009)、認知機能に関してはさらなる研究の継続が必要であると考えている。そこで本計画では、性成熟前期のMDマウスを様々な環境で飼育し、それに伴う認知機能の変化を研究することで、周産期ストレスが思春期の脳機能およびストレス脆弱性の形成に寄与するメカイズムを解明することを計画した。
本研究の結果、周産期ストレスを負荷した後のマウスを個別に飼育した場合と、集団で飼育した場合とで、その後の学習行動試験(タッチパネルオペラント装置を用いた視覚弁別課題)の結果に差が見られる可能性が示唆された。この原因について精査することを計画したが、研究代表者の異動・科研費申請資格喪失に伴い、研究終了となった。

In this study, we observed the role of astrocyte on remodeling for functional compensation in contralateral hemisphere of focal stroke. We found that dopamine is one of the strong candidate for contributing the functional remodeling/recovery after focal stroke. We also found that repetitive application of Aripiprazole, one of the partial agonist for dopamine receptor, enhances the remodeling of neuronal circuit in intact hemisphere after focal stroke. In this study, we observed the role of astrocyte on remodeling for functional compensation in contralateral hemisphere of focal stroke. We found that dopamine is one of the strong candidate for contributing the functional remodeling/recovery after focal stroke. We also found that repetitive application of Aripiprazole, one of the partial agonist for dopamine receptor, enhances the remodeling of neuronal circuit in intact hemisphere after focal stroke. Since the Aripiprazole enhances the activity of astrocyte if it repetitively applied, our study suggested that application of aripiprazole enhances the neuronal remodeling after focal stroke via activation of dopaminergic systems.

This study was designed to examine the effect of the alteration of nuclear receptor-mediated gene expression on brain development. Using in vivo preparation, we found that congenital hypothyroid mouse and rat showed cerebellar ataxia. Electrophysiological study showed that such alteration may be induced by an aberrant synaptic function between parallel fiber and Purkinje cell. Perinatal PCB treatment induced the similar phenotype. Using in vitro preparation, we established the primary culture system of the hippocampus to study the effect of thyroid hormone. In addition, we examined the affect of gadolinium-based contrast agents (GBCA) on thyroid hormone action and found that linear GBCA disrupted thyroid hormone receptor action and thyroid hormone-mediated cerebellar dendritogenesis of Purkinje cell.

母子乖離操作により慢性ストレスを付加したマウスを成体の時点で研究した結果、以下の知見を得た。
雄の母子乖離マウスにおいて、(1)母子乖離マウスでは体性感覚野における神経活動が亢進しており、それに相関して知覚閾値の低下がみられた。(2)同部位ではグルタミン酸の過剰放出が起こっていた。(3)ホームケージにいるときでもストレスホルモン(コルチコステロン)の分泌が増加していた。(4)急性ストレスの追加負荷により、通常みられるコルチコステロンの一過性上昇が、母子乖離マウスでは消失していた。一方、同時に計測したグルタミン酸の濃度は、母子乖離マウスで顕著に増加していた。(5)急性ストレス負荷後、対照群に比べ一部のグルタミン酸受容体の発現が過剰増加していた。(6)同マウスをin vivo imaging法により生きたまま観察すると、マイクログリアの動態が変化していることが分かった。以上の結果は2報の論文として執筆中である。

The neuronal K(+)-Cl(-) cotransporter (KCC2) is a membrane transport protein that extrudes Cl(-) from neurons and helps maintain low intracellular [Cl(-)] and hyperpolarizing GABAergic synaptic potentials. Depolarizing gamma-aminobutyric acid (GABA) responses in neonatal neurons and following various forms of neuronal injury are associated with reduced levels of KCC2 expression. Mutation to the putative tyrosine phosphorylation site within the long intracellular carboxyl terminus of KCC2(Y1087D) or application of the tyrosine kinase inhibitor genistein shifted the GABA reversal potential (E(GABA)) to more depolarized values, indicating reduced KCC2 function. This was associated with a change in the expression pattern of KCC2 from a punctate distribution to a more uniform distribution, suggesting that functional tyrosine-phosphorylated KCC2 forms clusters in restricted membrane domains. A tyrosine phosphatase inhibitor increased the proportion of KCC2 associated with lipid rafts membrane domains. These results indicate that direct tyrosine phosphorylation of KCC2 results in membrane clusters and functional transport activity. Oxidative stress resulted in a rapid dephosphorylation of KCC2 that preceded the decreases in KCC2 protein or mRNA expression. Dephosphorylation of KCC2 is correlated with a reduction of transport activity and a decrease in [Cl-]i, as well as a reduction in KCC2 surface expression. Manipulation of KCC2 tyrosine phosphorylation resulted in altered neuronal viability in response to in vitro oxidative stress. We propose that neuronal stress induces a rapid loss of tyrosine phosphorylation of KCC2 that results in translocation of the protein and functional loss of transport activity.
